Jesse Appleton was born 17 October 1772 in New Ipswich, Hillsborough, New Hampshire to Francis Appleton (1733-1816) and Elizabeth Hubbard (1730-1815) and died 12 November 1819 Brunswick, Cumberland County, Maine of unspecified causes. He married Elizabeth Means (1780-1844) 25 April 1800 in Boston, Suffolk County, Massachusetts.

After having graduated from Dartmouth College in 1792, Appleton taught at several institutions including Amherst College and then worked at a parish in Hampton, New Hampshire. In the early 19th century, he received the honorary degree of Doctor of Divinity from both Dartmouth and Harvard University.
